There are 30 main story missions in Marvel's Wolverine spread out over nine chapters. Most missions take around half an hour to complete, although some of them are a little shorter. 20 of the 30 missions contain collectibles, so you'll need to keep your eyes peeled as you make your way through them if you want to unlock everything.
A typical playthrough will last between 15 and 20 hours, but if you're still hungry for more once the credits have finished rolling, Marvel's Wolverine does have a New Game Plus mode that introduces new techniques and adaptations while increasing the level cap from 36 to 40, which should help subsequent playthroughs feel a bit different from the first.

While it may be possible to shave off a little bit of time by skipping collectibles and avoiding Nightmare Doors, the former tend to be out in the open, and Nightmare Trials can usually be completed in around a minute, so there really isn't too much difference between a completionist playthrough and a story-only run. Playing Marvel's Wolverine on a harder difficulty setting may increase your playtime a little, but not by much.
There are three main collectible types in Marvel's Wolverine. Nightmare Doors allow Wolverine to revisit his past trauma and can be found by following the purple tree roots that appear near their locations. After passing through a door, you'll need to complete a short trial, which will unlock a memory and up to 4 AP, depending on how quickly you complete the trial. Whiskey Bottles also award AP and can be found by following blue scent lines. Last but not least are Material Crates, which contain the materials that you'll need to craft new suits, and can be tracked down by following the audio cues from your controller and the sonar-like pings that they emit on-screen. Crafting new suits increases Wolverine's health, so tracking down Material Crates is something of a must if you're playing on one of the harder difficulty settings.
Don't worry if you end up missing a few collectibles here and there, as after completing your first Nightmare Trial in the Homecoming mission, you'll be able to replay completed missions by interacting with the bed in the Nightmare Cabin. You'll find yourself there whenever you complete a Nightmare Trial, but you can also access the Nightmare Cabin via the main menu once it's unlocked.
