While you're technically free to anger the court members in any order and explore Vale Sangora as you see fit, The Blood of Dawnwalker's map is clearly designed so that Coen will naturally encounter Ambrus' loyalists first.
Ambrus' activities are all fairly easy to complete, and you'll likely stumble upon things like transports without seeking them out. However, until you get close to some of these things, they'll likely only appear as question marks on the map. Once you complete them, Ambrus' meter will rise and so will your overall infamy.
Ambrus is surprisingly popular among people, at least compared to the other members of Brencis' court, so not as many people will be as willing to betray him as you might think. Ambrus' order of business in Vale Sangora seems to revolve around collecting leeches to keep blood fresh and stocking up on as much blood as he can by draining it from prisoners.
To anger him, you'll need to free prisoners, stop his men from collecting leeches, destroy barrels, and disrupt overall transport. At almost every location, there will be barrels to destroy to complete the task. Once you have sufficiently angered him, you'll receive a special quest tied to him.
You don't always have to wait for the quest to take out the members of Brencis' court, but the quests typically give additional content and unique scenes, so I'd recommend it if you have the time. With that said, each of these will require different amounts of time to complete.
You do not need to complete all of these to max out his anger meter, but you can complete them after he's dead to increase your overall infamy.
For these activities, you'll need to destroy the blood supply that Ambrus has stored and defeat all the guards.
